Abstract: Neste trabalho, foi realizada a caracterização molecular do gene da β-tubulina de Phakopsora pachyrhizi e da proteína codificada por este gene. Uredósporos foram coletados em sete municípios brasileiros. O DNA genômico foi extraído e o gene da β-tubulina amplificado pela técnica de PCR, por meio de cinco combinações de primers específicos de modo a cobrir toda a região desejada. Os fragmentos obtidos foram purificados e sequenciados. As sequências foram alinhadas e comparadas entre si e com sequências obtidas no GenBank. O gene da β-tubulina de P. pachyrhizi apresentou 2815 pares de bases, sendo constituído de nove éxons e oito íntrons. Sua fase aberta de leitura (ORF) apresentou 1341 pares de bases. Verificou-se que 452 pares de bases são correspondentes à região promotora, enquanto 234 pares de bases pertencem à extremidade 3 não traduzida. A região promotora apresentou os sinais típicos envolvidos com o início da transcrição, sendo três possíveis sequências TATA box e cinco possíveis sequências CAAT box. Um domínio de 30 nucleotídeos, constituído por citosina e timina, está contido na região promotora entre os pares de bases -205 e -175 upstream ao códon de inicio da tradução. Sítios envolvidos no mecanismo de splicing estão presentes, flanqueando os oito íntrons, sendo o sítio GT(A/G)(N)GT encontrado na extremidade 5 do íntron e o sítio (C/T)AG na extremidade 3 . A sequência consenso AATAAA, que desempenha papel importante no processo de poliadenilação do mRNA precursor, foi encontrada a 44 pares de bases downstream do códon de terminação TAG. A ORF encontrada para este gene codifica uma proteína constituída por 446 aminoácidos, a qual apresenta elevada identidade (acima de 89%) com β-tubulina de outras espécies de fungos relacionados. Ao comparar a sequência de aminoácidos predita para β-tubulina, verificou-se que esta proteína possui quatro domínios conservados, sendo eles: domínio de ligação a nucleotídeos, domínio de ligação ao taxol, domínio de interface alfa/beta e domínio de interface beta/alfa. Análises filogenéticas, com base no alinhamento do gene da β-tubulina, proporcionaram a alocação de P. pachyrhizi junto aos demais fungos causadores de ferrugens. Comparação dos íntrons do gene da β-tubulina de P. pachyrhizi com íntrons de Melampsora lini e Uromyces fabae, sendo as duas últimas as únicas espécies de fungos causadores de ferrugens com o gene da β-tubulina completamente sequenciado até o momento, mostrou conservação no número e posição dos íntrons. Entretanto, as sequências correspondentes a cada um dos íntrons mostraram baixa identidade (aproximadamente 30%).
This study was carried out to characterize either the β- tubulin gene from the Phakopsora pachyrhizi and the protein codified by this gene. The urediniospores were collected in seven Brazilian counties. The genomic DNA was extracted and the β-tubulin gene was amplified by PCR technique through five combinations of specific primers on such a way to covering the whole region under study. Then, the obtained fragments were purified and sequenced. The sequences were aligned and compared among each others and with sequences obtained in the GenBank. The β-tubulin gene from P. pachyrhizi showed 2815 pairs of bases and was constituted by nine exons and eight introns. Its opened reading phase (ORF) showed 1341 base pairs. It was verified that 452 base pairs correspond to the promoter region, whereas 234 pairs belong to the untranslated extremity 3'. The promoter region showed the typical signs involved in the beginning of the transcription, as being three possible sequencies TATA box and five possible sequencies CAAT box. A domain of 30 nucleotides constituted by cytosine and thymine is inside the promotor region between the basis pairs - 205 and -175 upstream the codon beginning the translation. Some sites involved in the splicing mechanism are found, as flanking those eight introns, whereas the site GT(A/G)(N)GT was found at the extremity 5' of the intron and the site (C/T)AG at extremity 3'. The consensus sequence AATAAA that plays an important role in the polyadenilation process of the precursory mRNA was found after 44 pairs of bases downstream the TAG termination codon. The ORF found for this gene codifies a protein constituted by 446 amino acids, that shows high identity (above 89%) with β-tubulin in other fungus species related. When comparing the amino acid sequence predicted for β-tubulin, this protein was identified to have four conserved domains as follows: domain of linkage to nucleotides, domain of linkage to taxol, domain of alpha/beta interface and domain of beta/ alpha interface. The phylogenetic analyses based on alignment of the β-tubulin gene provided the allocation of P. pachyrhizi together with the other fungus causing the rusts. The comparison of the introns of the β-tubulin gene of P. pachyrhizi with introns of Melampsora lini and Uromyces fabae, as the two last ones being the only rustcausing fungus species with the β-tubulin gene completely sequenced until the moment, showed that both number and position of the introns were conserved. However, the sequences corresponding to each intron showed low identity (around 30%).
