Abstract: Este estudo objetivou avaliar os efeitos da criopreservação pelo método convencional em ovócitos imaturos e maturados in vitro. O experimento foi conduzido no Laboratório de Reprodução Animal - DVT-UFV, utilizando-se ovócitos provenientes de ovários de vacas abatidas em matadouro e distribuídos aleatoriamente em seis tratamentos. Tratamento 1 (T1): ovócitos não congelados (frescos) providos de células do cumulus oophorus (COC), os quais foram imediatamente submetidos à MIV, FIV e CIV. Tratamento 2 (T2): ovócitos não congelados (frescos) e desprovidos de células do cumulus oophorus (desnudados), os quais foram imediatamente submetidos ao MIV, FIV e CIV. Tratamento 3 (T3): ovócitos imaturos, providos de células do cumulus oophorus, submetidos à criopreservação imediatamente após a seleção. Posteriormente, esses ovócitos foram descongelados e aqueles considerados normais foram submetidos à MIV, FIV e CIV. Tratamento 4 (T4): ovócitos desnudados e submetidos à criopreservação. Posteriormente, foram descongelados e aqueles considerados normais foram submetidos à MIV, FIV e CIV. Tratamento 5 (T5): ovócitos providos de células do cumulus oophorus, maturados in vitro e, em seguida, congelados. Posteriormente, foram descongelados e aqueles considerados normais foram submetidos ao recultivo em meio de maturação por duas horas e, então, submetidos à FIV e CIV. Tratamento 6 (T6): ovócitos desnudos, maturados in vitro e, posteriormente, submetidos a criopreservação. Após a descongelação, os considerados normais foram submetidos ao recultivo por duas horas seguido de FIV e CIV. A congelação dos ovócitos dos tratamentos 3, 4, 5 e 6 foi realizada em solução contendo 1,8 mol L-1 de etileno glicol (EG) em Talp-Hepes (meio base) acrescido de 0,4% de BSA-V, utilizando-se o método convencional. Os ovócitos foram desidratados por imersão em três soluções (etapas), contendo concentrações crescentes do crioprotetor, (0,6; 1,2 e 1,8 mol L-1 de EG) sendo o tempo de permanência máxima de cinco minutos em cada etapa, em temperatura ambiente. A descongelação foi realizada por imersão em banho-Maria a 30oC por 20 segundos. Posteriormente, os ovócitos foram reidratados em três etapas (0,9 mol L-1 de EG + 0,3 mol L 1 de sacarose; 0,3 mol L-1 de sacarose, e sem EG e sem sacarose), com duração de seis minutos cada uma. Após a descongelação foram recuperados 92,6, 97,5, 96,6 e 93,2%, dos ovócitos dos tratamentos 3, 4, 5 e 6, respectivamente. Em relação aos ovócitos recuperados, verificou-se uma taxa de citoplasma retraído de 3,3; 0,8; 0,3 e 6,2% e perda de conteúdo celular de 2,2; 13,1; 3,8 e 16,1%, também para os tratamentos 3, 4, 5 e 6, respectivamente. Os ovócitos congelados imaturos apresentaram taxa de maturação de 9,2 e 5,8% (tratamentos 3 e 4, respectivamente), resultados estes bem inferiores aos obtidos para os ovócitos frescos, sendo 82,5 (T1) e 75,4% (T2). Foram encontradas taxas de fecundação de 56,2; 0,0; 38,7; 8,6; 63,6 e 16,7% e de clivagem de 36,3; 7,9; 0,4; 0,0; 0,0 e 0,0%, para os tratamentos 1, 2, 3, 4, 5 e 6, respectivamente. Somente os ovócitos não congelados e não desnudados (T1) apresentaram desenvolvimento para mórulas e blastocistos (34,5%), após a fecundação. Estes resultados indicam que o protocolo de congelação adotado comprometeu a viabilidade do ovócito.
This study aimed at the evaluation of the effects from cryopreservation on the in vitro matured and immature oocytes, using the conventional method. The experiment was carried out in the Animal Reproduction Laboratory - DVT-UFV, by using oocytes from cows ovaries from slaughterhouse, which were distributed into six treatments. Treatment 1 (T1): non-frozen oocytes (fresh ones) provided with the cumulus oophorus cells (COC) which were immediately submitted to the processes MIV, FIV and CIV. Treatment 2 (T2): non-frozen oocytes (fresh ones), deprived unprovided of the cumulus oophorus cells (naked), which were immediately submitted to MIV, FIV and CIV. Treatment 3 (T3): immature oocytes provided with the cumulus oophorus cells, which were submitted to cryopreservation just after selection. Later, they were thawed and those considered as normal ones were submitted to MIV, FIV and CIV. Treatment 4 (T4): naked oocytes submitted to cryopreservation. Later, they were thawed and those considered as normal ones were submitted to MIV, FIV and CIV. Treatment 5 (T5): oocytes provided with the cumulus oophorus cells, matured in vitro, and then they were frozen. Later, they were thawed and those considered as normal ones were submitted to recultivation for 2 hours in a maturation medium, and then submitted to FIV and CIV. Treatment 6 (T6): naked, in vitro matured oocytes which were submitted to cryopreservation. After thawing, those oocytes which were considered as normal ones were submitted to recultivation for 2 hours, and then to FIV and CIV. Those oocytes of the treatments 3, 4, 5 and 6 were frozen in a solution containing 1.8 mol L-1 ethylene glycol (EG) in Talp-Hepes (base medium) added with 0.4% BSA-V by the conventional method. The oocytes were dehydrated by immersion into three solutions (phases) at increasing concentrations of the cryoproctetor, (0.6; 1.2 and 1.8 mol L-1 of EG) during a maximum permanence time of five minutes for each phase, at environmental temperature. Thawing was accomplished by immersion into water bath at 30oC for 20 seconds. Later, the oocytes were rehydrated at three stages, that is, 0.9 mol L-1 EG + 0.3 mol L -1 sucrose, 0.3 mol L-1 sucrose, and without both EG and sucrose for six minutes each one. After thawing, the oocytes recovery rates were 92.6, 97.5, 96.6 and 93,2% for the treatments 3, 4, 5 and 6, respectively. In relation to the recovered oocytes, the following were also found: a cytoplasm withdrawal rate of 3.3, 0.8, 0.3 and 6.2%, and a cellular content loss of 2.2, 13.1, 3.8 and 16.1%, also for treatments 3, 4, 5 and 6, respectively. The frozen immature oocytes presented a maturation rate of 9.2 and 5.8% (treatments 3 and 4, respectively); these results were much inferior to those obtained by the fresh oocytes, that is, 82.5 (T1) and 75.4% (T2). The fecundation rates were 56.2, 0.0, 38.7, 8.6, 63.6 and 16.7%, while the clivage rates were 36.3, 7.9, 0.4, 0.0, 0.0, and 0.0% for treatments 1, 2, 3, 4, 5 and 6, respectively. Only the unfrozen and no-naked oocytes (T1) exhibited development of morula and blastocyst (34.5%), after fecundation. These results point out that the adopted freezing protocols affected the viability of the oocytes.
