Abstract: Leveduras floculantes são de grande utilidade em processos fermentativos, pois são separadas facilmente do mosto de fermentação. Diminuindo os custos associados á etapa de separação das células. O fenótipo de floculação não é encontrado com frequência em leveduras nativas, incluindo a espécie Kluyveromyces marxianus a qual apresenta alto potencial biotecnológico devido a sua hablilidade de assimilar um amplo espectro de substratos, apresentar uma alta taxa de crescimento e ser termotolerante. Este trabalho teve como objetivo a construção de linhagens floculantes de K. marxianus com potencial para fermentar resíduos agroindustriais. Os cassetes utilizados na construção das linhagens floculantes de K. marxinaus foram amplificados de 4 linhagens floculantes de Saccharomyces cerevisiae que possuem, cada uma, um cassete constituído pelo o gene URA3 como marca de seleção e por um gene de floculação FLO, sob o controle do promotor constitutivo TDH3 (URA3-TDH3p-FLO). Sendo que cada uma dessas linhagens contém um gene de floculação diferente (FLO1, FLO5, FLO9 E FLO10). A fim de obter um mutante auxotrófico da linhagem de K. marxinaus UFV-3 o gene URA3 desta linhagem foi amplificado, clonado e um fragmento da sua região codificadora foi retirado obtendo-se um gene inativo URA3Δ que foi utilizado na transformação da K. marxinaus UFV-3. Neste trabalho não foi possível obter uma linhagem de K. marxinaus UFV-3 auxotrófica para uracila, por isso utilizamos a linhagem CBS 6556 já auxotrófica para uracila. Esta linhagem foi transformada por eletroporação utilizando os cassetes FLO obtendo-se 4 diferentes linhagens floculantes de K. marxianus CBS 6556 que foram selecionadas em meio sem uracila. A análise fenotípica dos transformantes mostrou que que todas as linhagens eram floculantes, contudo, o grau de floculação variou entre as 4 linhagens. Avaliou-se o perfil fermentativo das linhagens floculantes de K. marxianus utilizando glicose, lactose e glicose mais lactose como fonte de carbono e energia. Foi verificado que a linhagem transformada com o cassete contendo o gene FLO9 (KMCBSFLO9) apresentou melhor capacidade fermentativa e manteve sua capacidade de floculação. Com a finalidade de verificar a influência do fenótipo de floculação na viabilidade celular em resposta ao etanol a linhagem de KMCBSFLO9, assim como da linhagem menos floculante (KMCBSFLO10), e a não floculante (KMCBSΔURA3) foram cultivadas na presença de diferentes concentrações de etanol. Os resultados demonstraram que tanto a linhagem não floculante como as duas linahgens floculantes se mostraram resistentes ao etanol.
Flocculent yeasts are useful in fermentation processes because they can be easily separated from the fermentation mash reducing costs with cell separation equipment. The flocculation phenotype is not often found in native yeasts, including the species Kluyveromyces marxianus which has a high biotechnological potential due to its ability to assimilate a variety of substrates, ferment at higher temperatures and present a high growth rate. This work aimed at the construction of flocculent strains of K. marxianus with potential to ferment agro-industrial residues. The cassettes used in the construction of flocculent strains of K. marxinaus were amplified from 4 different flocculent strains of Saccharomyces cerevisiae, each strain has a cassette consisting of the URA3 gene as a selection mark and a flocculation gene FLO under control of the constitutive promoter TDH3 (URA3-TDH3p-FLO) and each of these strains contains a different flocculation gene (FLO1, FLO5, FLO9 and FLO10). In order to obtain an auxotrophic mutant strain of K. marxinaus UFV-3, the URA3 gene of this strain was amplified and cloned. Then a fragment of its coding region was removed yielding an inactive gene (URA3Δ), which was used in the transformation of K. marxinaus UFV-3. Due to the impossibility to obtain an auxotrophic strain of K. marxinaus UFV-3, we used the CBS 6556 strain auxotrophic for uracil. This strain was transformed by electroporation using the cassettes from S. cerevisiae. We obtained 4 flocculent strains of K. marxianus CBS 6556 that were selected by growth in medium without uracil. By carrying out the phenotypic analysis of the transformants, it was found that all the strains were flocculants; however, the flocculation degree was different between the 4 strains. The fermentation characteristics of the flocculent strains of K. marxianus were evaluated by using glucose, lactose and glucose/lactose as carbon and energy source and it was found that the strain transformed with the cassette containing the gene FLO9 (KMCBSFLO9) was the strain that showed the best fermentative performance preserving its ability to flocculation. In order to verify the influence of flocculation phenotype in the alcohol tolerance, the KMCBSFLO9 strain, the less flocculant strain (KMCBSFLO10) and the no-flocculant strain (KMCBSΔURA3) were cultured in the presence of different concentrations of ethanol. The results showed that the flocculent and no-flocculant strains present higher cell viability after 12 hours of culture in the presence of different concentrations of ethanol.
