Título: Identificação e capacidade de adesão de Staphylococcus spp. isolados de manipuladores, superfícies e ar de ambientes de uma indústria de laticínios
Identification and the ability to adhere of Staphylococcus spp. species isolated of the manipulators hands, food contact surfaces and environmental air in a dairy plant

Abstract: O objetivo deste trabalho foi determinar a ocorrência de S taphylococcus spp. no ar, nas superfícies de equipamentos e nas mãos de manipuladores de ambientes de recepção, pasteurização, embalagem e produção de doce de leite, manteiga, queijos e iogurtes de um laticínio. A técnica de impressão em ágar foi empregada para quantificar o grau de contaminação do ar. A técnica de swab foi empregada para a contagem desses microrganismos nas superfícies e nas mãos dos manipuladores. Estafilococos isolados dos ambientes foram identificados e submetidos ao teste de produção e identificação de enterotoxinas SET-RPLA (OXOID ®) dos tipos A, B, C e D. A capacidade de adesão de quatro espécies de estafilococos produtores e pauciprodutores de enterotoxinas foi avaliada em superfícies de polipropileno, polietileno de baixa densidade, aço inoxidável AISI 304 #4 e vidro, por meio de microscopia de epifluorescência. Não foi verificada diferença significativa (p>0,05) entre as médias do logaritmo do número médio de Staphylococcus spp. para o ar dos ambientes, cujo número atingiu 0,6 log UFC.m-3. Essa diferença também não foi constatada para superfícies (1,01 log UFC.cm-2) e para mãos dos manipuladores (1,83 log UFC.mão-1). Dos 137 isolados de Staphylococcus spp., 46 foram provenientes do ar, 40 das superfícies e 51 das mãos dos manipuladores. As espécies mais freqüentes, acima de 10% do total de isolados, no ar e nas superfícies foram S. xylosus, S. lentus e S. aureus. Os isolados das mãos dos manipuladores, de S. epidermidis, além de S. aureus e S. xylosus, foram também identificados com porcentual acima de 10%. Dezessete isolados (12,4%) produziram pelo menos um dos tipos de enterotoxina. Isolados coagulase negativa de S. haemolyticus, S. sciuri e S. saprophyticus provenientes do ar produziram toxinas A, B C e D; entretanto, S. epidermidis produziu os tipos A e D, e uma outra estirpe produziu os tipos A, C e D. As espécies S. lugdunenensis, S. capitis e S. xylosus, isoladas de equipamentos, também coagulase negativa, produziram todos os tipos de toxina. S. hominis, isolado das mãos dos manipuladores, produziu as toxinas A, B e C, enquanto o isolado de S. capitis produziu os quatro tipos. Dentre os três isolados de S. epidermidis, um não produziu a toxina tipo C, e as demais produziram os tipos A, B, C e D. Para os estudos de adesão selecionaram-se S. epidermidis, isolado do ar do ambiente , e S. xylosus, isolado da superfície dos equipamentos, ambos produtores das quatro enterotoxinas, além de dois isolados das mãos dos manipuladores; um deles, S. saprophyticus, produtor de quatro enterotoxinas, e o outro, S. aureus, não-produtor de toxina. Observou-se que os isolados aderiram a todas as superfícies avaliadas. Não houve diferença significativa (p>0,05) no grau de adesão para as superfícies, constatando-se médias de 5,75 log UFC.cm-2 e 5,68 log UFC.cm-2, 5,87 log UFC.cm-2 e 5,84 log UFC.cm-2 para polipropileno, polietileno de baixa densidade, aço inoxidável ASI 304 #4 e vidro, respectivamente. Houve diferença significativa (p<0,05) quanto ao grau de adesão dos microrganismos avaliados. S. epidermidis apresentou menor capacidade de adesão em polietileno de baixa densidade (5,16 log UFC.cm-2) e os isolados S. aureus (5,88 log UFC.cm-2), S. xylosus (5,97 log UFC.cm-2) e S. saprophyticus (5,72 log UFC.cm-2) não diferiram entre si.
Staphylococcus spp. isolated in the air, in the food contact surfaces and in the manipulators at processing areas in a dairy plant, including milk reception, packaging, and pasteurization rooms, rooms where cheese, yogurt, butter and “doce de leite” (Latin American typical treat made of concentrated milk and sugar), were evaluated. The impression technique in agar using air sampler was used to quantify the degree of contamination of the air, in UFC.m-3. For the swab technique was determined the counting of those microorganisms in the surfaces, expressed in UFC.cm-2 and in the manipulators expressed in UFC/hand. The staphylococci isolated of air were submitted the biochemical identification by the system API Staph (BIOMERIEUX®) and the to the production and identification of enterotoxins test SET-RPLA (OXOID®), that it detects the types A, B, C and D of the enterotoxins. Also, It was evaluated the ability of adhesion of four species of producing staphylococci and low- enterotoxin-producing, based in their identification with biochemical characteristics, above 90% degree of identification, in polypropylene, low density polyethylene, stainless steel AISI 304 #4 and glass surfaces, by using epifluorecence microscopy (LEICA DC 300F). There were not differences (p>0.05) between the averages of the logarithm of the number of Staphylococcus spp. for the air, with average of 0.6 log UFC.m-3. That difference, also, was not significant for surfaces (1.01 log UFC.cm-2) and for manipulators (1.83 log UFC.mão-1). For the 137 isolated of Staphylococcus spp., only 46 were from the air, 40 of the surfaces and 51 of the manipulators. Above 10%, the most frequent species, in the air and in the surfaces they were S. xylosus, S. lentus and S. aureus. For manipulators, the species S. epidermidis, besides S. aureus and S. xylosus were also identified with percentage above 10%. Seventeen isolated (12.4%) produced at least one of the enterotoxin types. In the air, the isolated negative coagulase S. haemolyticus, S. sciuri and S. saprophyticus produced toxins A, B C and D. The S. epidermidis produced the types A and D and another strains produced the types A, C, D. For the surfaces of equipments, the species S. lugdunenensis, S. capitis and S. xylosus also negative coagulase produced all of the toxin types. In the manipulators' hands, to the isolated identified like S. hominis produced three types of toxins A, B, C while isolated S. capitis produced the four identified types for the method. By the way three of the isolated of S. epidermidis, only one did not produce the toxin type C, while the others produced the types A, B, C and D. For the adhesion studies they were selected isolated S. epidermidis obtained of the atmosphere air, S. xylosus originating from surface of equipments, both producing four enterotoxinas types, besides isolated from manipulators; one was S. saprophyticus, producing of four enterotoxins and another was S. aureus no toxin producer. All the isolated adhered to the surfaces evaluated. There was not difference (p>0.05) for the adhesion in the surface e evaluated with averages of 5.75 log UFC.cm-2 and 5.68 log UFC.cm-2, 5.87 log UFC.cm-2 and 5.84 log UFC.cm-2 for polypropylene, polyethylene of low density, stainless steel ASI 304 #4 and glass, respectively. There was difference (p<0.05) in the adhesion of the microorganisms evaluated. S. epidermidis presented lower adhesion capacity in polyethylene of low density (5.16 log UFC.cm-2), isolated S. aureus (5.88 log UFC.cm-2) and S. xylosus (5.97 log UFC.cm-2) and S. saprophyticus (5.72 log UFC.cm-2), they did not differ amongst themselves.
