Título: Micro-organismos Eficientes: diversidade microbiana e efeito na germinação, crescimento e composição química de capim-marandu
Effective microorganisms: microbial diversity and effect on germination, growth and chemical composition of palisade grass

Abstract: Os Micro-organismos Eficientes (EM), inoculantes formados por fungos e bactérias isolados de matas, podem influenciar a germinação de sementes e o crescimento de plantas. Não foram encontrados trabalhos identificando a composição microbiana desses inoculantes, assim, foram realizados dois experimentos com os seguintes objetivos: Experimento 1: analisar, pela técnica de PCR-DGGE, o perfil da comunidade microbiana em três inoculantes EMs de três origens, a eficiência no crescimento e na composição química de capim-marandu, cultivado em solo acrescido ou não de esterco bovino, e o perfil microbiano desse inoculante no ambiente de cultivo; Experimento 2: analisar a diversidade por sequenciamento Illumina MiSeq e caracterizar taxonomicamente fungos e bactérias de três inoculantes EM de origens distintas, além de avaliar a respectiva eficiência na germinação de sementes de capim-marandu. No experimento 1, as plantas foram cultivadas em vasos mantidos em casa de vegetação, em esquema fatorial 2 x 4, com cinco repetições, consistindo de solo sem ou com esterco bovino e três tipos de EM (EM1, EM2 e EM3) + controle sem inoculação, totalizando 40 unidades experimentais. O EM1 é de origem comercial e os outros dois de origem caseira, produzidos na região de Muriaé (EM2) e Viçosa (EM3), ambas localizadas na Zona da Mata de Minas Gerais. Nos tratamentos que receberam o esterco foram adicionados 90 g deste na superfície do solo e naqueles que receberam EM foram realizadas três aplicações de 34 mL, a cada 30 dias. Aos 60 e 100 dias de experimento foram avaliadas características de crescimento e químicas do capim-marandu. O perfil de bactérias e fungos dos inoculantes EM e do solo foi obtido conforme a técnica PCR-DGGE. No experimento 2, o DNA dos três inoculantes EM foi extraído, seguido de amplificação por PCR do gene rRNA 16S e da região ITS de fungos e do sequenciamento na plataforma Illumina MiSeq. Testes de germinação foram conduzidos no delineamento Inteiramente Casualizado (DIC), com esquema fatorial 3 x 3 x 2 + 1 (controle positivo) + 2 (controles negativos) e quatro repetições de 50 sementes. Os tratamentos consistiram em: pré-tratamentos com três tipos de EM (EM•1®, EM 2 e EM3) em três concentrações (1% em água; 2% em água e 100% de EM) e dois tempos de imersão (5 min e 24 h); pré-tratamentos com ácido sulfúrico 18% por 15 min e com água (5 min e 24 h). Houve maior crescimento das plantas nos tratamentos com esterco. Na ausência de esterco o EM2 foi destaque por melhorar algumas características das plantas, incluindo comprimento da parte aérea (CPA) e diâmetro do colmo (DC) no primeiro corte e CPA no segundo corte. Nos tratamentos com o esterco, as características agronômicas, analisadas aos 60 e 100 dias de experimento, não foram afetadas, entretanto, houve pronunciado aumento no teor de proteína bruta (PB) ao utilizar o EM2. Dependendo da origem, inoculantes EM diferem quanto a estrutura da comunidade de fungos e bactérias. A presença e a ausência de esterco no solo determinaram o perfil bacteriano e fúngico, com efeito pronunciado na comunidade fúngica dos solos com EMs 1 e 2. O grupo de bactérias foi o mais abundante nos EMs, seguido pelos fungos. EMs 2 e 3 não compartilharam unidades taxonômicas operacionais (OTUs) de fungos, no entanto, algumas UTOs bacterianas foram compartilhadas por todos os EMs. Muitas dessas bactérias são promotoras de crescimento vegetal e produzem fitormônios, o que pode justificar a eficiência destes inoculantes na germinação de sementes. Pré-tratamentos de sementes de capim-marandu com os três EMs resultaram em maior porcentagem de germinação (%G) e índice de velocidade de germinação (IVG) quando o EM foi utilizado na concentração 1 e 2%. A imersão das sementes por 5 min foi mais eficiente comparada a imersão por 24h. A %G e o IVG das sementes tratadas com ácido sulfúrico não diferiram dos tratamentos com EM, mas ambos foram superiores ao controle.
Effective Microorganisms (EM), inoculant formed by fungi and bacteria isolated from forests, can influence seed germination and plant growth. No experiments were found to identify the microbial composition of these inoculants, so two experiments were carried out with the following objectives: Experiment 1: to analyze by the PCR-DGGE technique the profile of the microbial community in three inoculants EMs from three origins, the efficiency in the growth and chemical composition of palisade grass, cultivated in soil with or without bovine manure, and the microbial profile of this inoculant in the growing environment; Experiment 2: to analyze diversity by Illumina MiSeq sequencing and taxonomically characterize fungi and bacteria of three EM inoculants of different origins, in addition to evaluating their efficiency in the germination of palisade grass seeds. In experiment 1, the plants were grown in pots kept in a greenhouse, in a 2 x 4 factorial scheme, with five replicates, consisting of soil without or with bovine manure and three types of EM (EM1, EM2 and EM3) + control without inoculation, totalizing 40 experimental units. EM1 is of commercial origin and the other two of home origin, produced in the region of Muriaé (EM2) and Viçosa (EM3), both located in the Zona da Mata of Minas Gerais. In the treatments that received manure, 90 g of this was added to the soil surface and in those receiving EM, three 34 mL applications were performed every 30 days. At 60 and 100 days of experiment were evaluated growth and chemical characteristics of the palisade grass. The bacteria and fungi profiles of the EM and soil inoculants were obtained according to the PCR-DGGE technique. In experiment 2, DNA from the three EM inoculants was extracted, followed by PCR amplification of the 16S rRNA gene and the ITS region of fungi and sequencing on the Illumina MiSeq platform. Germination tests were conducted in a completely randomized design (CRD), with factorial scheme 3 x 3 x 2 + 1 (positive control) + 2 (negative controls) and four replicates of 50 seeds. The treatments consisted of: pre-treatments with three types of EM (EM•1®, EM2 and EM3) in three concentrations (1% in water, 2% in water and 100% EM) and two immersion times (5 min and 24 h); Pre-treatments with 18% sulfuric acid for 15 min and with water (5 min and 24 h). There was higher plant growth in manure treatments. In the absence of manure EM2 was highlighted by improving some plant characteristics, including shoot length (SL) and stalk diameter (SD) in the first cut and SL in the second cut. In the treatments with the manure, the agronomic characteristics, analyzed at 60 and 100 days of experiment, were not affected, however, there was a pronounced increase in crude protein (CP) content when using EM2. Depending on the origin, inoculants EM differ in the community structure of fungi and bacteria. The presence and absence of manure in the soil determined the bacterial and fungal profile, with a pronounced effect on the fungal community of soils with EMs 1 and 2. The group of bacteria was the most abundant in the EM, followed by fungi. EMs 2 and 3 did not share fungus operative taxonomic units (OTUs), however, some bacterial UTOs were shared by all EMs. Many of these bacteria are promoters of plant growth and produce phytonutrients, which may justify the efficiency of these inoculants in seed germination. Pre-treatments of palisade grass seed with the three EM resulted in a higher percentage of germination (%G) and germination speed index (GSI) when EM was used in the concentration 1 and 2%. The seed immersion for 5 min was more efficient compared to immersion for 24 hours. The %G and GSI of the seeds treated with sulfuric acid did not differ from the EM treatments, but both were superior to the control.
