Abstract: O presente trabalho teve por objetivo o estudo da influência de diferentes vedações na germinação in vitro, o estabelecimento de um protocolo de regeneração in vitro de explantes radiculares, a implementação de um protocolo de transformação genética mediada por Agrobacterium rhizogenes e A. tumefaciens, além de testar diferentes concentrações do surfactante Pluronic® F68 na regeneração de duas espécies de maracujazeiros (Passiflora cincinnata Masters e P. edulis Sims f. flavicarpa Degener. Quanto a germinação in vitro, os sistemas de vedação mais efetivos para proporcionar maior comprimento de hipocótilos e de raízes foram as embalagens sanfonadas de polipropileno com filtro microbiano e placa de Petri com fita hipoalergênica, por promoverem trocas gasosas mais efetivas e conseguinte produção de plântulas mais vigorosas. A organogênese em explantes radiculares em P. cincinnata e P.edulis f. flavicarpa ocorreu pela via direta, confirmada por estudos anatômicos e pela microscopia eletrônica de varredura. Ficou evidenciada a diferenciação de brotações a partir das células do câmbio vascular com intensas divisões celulares no plano periclinal e com típica conexão vascular com os explantes. As respostas dos genótipos estudados ocorreu em épocas diferentes, sendo um processo assincrônico no qual a suplementação do regulador de crescimento 6-benzilaminopurina (BA) ao meio de cultivo é indispensável para a resposta organogênica de explantes radiculares. Brotações induzidas em meio semi-sólido e alongadas em meio líquido sob agitação foram enraizadas em substrato de fibra de coco e Plantmax® (1:1), e aclimatizadas com sucesso. A citometria de fluxo indicou que não houve variação na quantidade de DNA dos regenerantes aclimatizados de todos os genótipos. A quantidade de DNA (pg) nos regenerantes de P. cincinnata (2,99 pg) e P. edulis f. flavicarpa (3,26- 3,28 pg) foram compatíveis com as quantidades de DNA das plantas controle, obtidas por via seminífera. Transformação genética de maracujazeiro utilizando Agrobacterium rhizogenes R1601 carregando a construção quimérica nos-nptII- xiiinos, o qual confere resistência à canamicina, e A. tumefaciens GV3101 carregando o gene APETALA com construção quimérica pROKII-AP1-GUSint, conferindo resistência à higromicina. Para a eliminação das agrobactérias após a fase de pré e co-cultivo, os antibióticos Meropenem e Cefotaxima foram mais efetivos na eliminação das bactérias, não interferindo na regeneração adventícia. O diagnóstico de PCR confirmou a presença do T-DNA e Ri-TDNA em todos os transformantes de P. cincinnata e P. edulis f. flavicarpa. Esse sistema mostra-se promissor para o estudo de genes com funções desconhecidas, bem como o estudo dos genes envolvidos na via da arquitetura floral, e também a facilidade de manipulação das raízes hairy root e a possibilidade de regeneração adventícia de brotações e de plantas transformadas. A utilização de Pluronic® F-68 na regeneração de brotos mostrou-se eficiente, sendo que para a espécie P. cincinnata as concentrações mais eficientes foram as mais baixas do surfactante (0,001 e 0,01%) enquanto que para P. edulis f. flavicarpa a resposta variou entre os genótipos estudados.
The present work aimed to study the influence of different type of sealing material on the in vitro germination, the establishment of a protocol for root explants in vitro regeneration, the implementation of a Agrobacterium rhizogenes and A.tumefaciens-mediated transformation protocol, besides to test different concentrations of the surfactant Pluronic® F68 in the regeneration of two passionfruit species (Passiflora cincinnata Masters and P. edulis Sims f. flavicarpa Degener genotypes FB 100, FB 200 and FB 300. The most effective systems of sealing material to provide greater hypocotyls and roots length were the concertina packaging of polypropylene with microbial filter and the petri dish with hypoallergenic tape, by promoting more effective gas exchange and therefore produce more vigorous seedlings. The organogenesis in P. cincinnata and P.edulis f. flavicarpa root explants occurred through direct way, confirmed by anatomical studies and scanning electron microscopy.It evidenced the differentiation of shoots from the the vascular cambium cells with intense periclinal cell divisions and with typical vascular connection with the explants. The responses of the studied genotypes occurred at different periods, and is an asynchronous process in which the supplementation of 6-benzylaminopurine (BA) plant growth regulator to the culture medium is essential for the organogenic response of root explants. Induced shoots in semi-solid medium and elongated in liquid medium under agitation were rooted in coconut-fiber substrate and Plantmax® (1:1), and successfully acclimatized. Flow cytometry indicated that there was no variation in the DNA amount of acclimatized regenerating of all genotypes. The DNA amount (pg) in regenerating of P. cincinnata (2.99 pg) and P. edulis f. flavicarpa (3,26-3,28 pg) were consistent with the DNA amounts from control plants, obtained by seminiferous via. The protocol presented can be useful as explants alternative for use in Agrobacterium-mediated genetic transformation of passionfruit. In the genetic transformation using A. rhizogenes R1601 carrying the chimeric construction xvnos-nptII-nos, which confers resistance to kanamycin, and A. tumefaciens GV3101 carrying the gene APETALA with the chimeric construction pROKII- AP1-GUSint, confering resistance to higromicina, for the Agrobacterium elimination after the phase of pre-and co-cultivation, the antibiotics Meropenem and cefotaxime were more effective, not interfering with the adventitia regeneration. The PCR diagnosis confirmed the presence of T-DNA and Ri- TDNA in all transformants of P. cincinnata and P. edulis f. flavicarpa. This system seems to be promising for the study of genes with unknown functions, as well as the study of genes involved in the floral architecture pathway, and also the facility of the roots "hairy root" handling and the possibility of adventitious shoots regeneration of the transformed plants. The use of Pluronic ® F-68 in the shoots regeneration was efficient, whereas for the P. cincinnata species most effective concentrations were the lowest ones (0.001 and 0.01%) while for P. edulis f. flavicarpa the response varied among the studied genotypes. The answers differ statistically regarding the control, being the most effective response for the P. edulis f. flavicarpa species the concentration of (0.01 and 0.1%). So the addition of Pluronic ® F-68 was effective in shoots induction for the studied species.
