Abstract: O ácido ferúlico é um intermediário da síntese de siringil e guaiacil, os monolignóis mais abundantes formadores da lignina. Além disso, participa da síntese da suberina e cutícula, atuando também como intermediário na síntese de alguns flavonóides. Por meio de análises posteriores em mutantes do gene OsABCB17, um gene da família ABC, foi verificado a existência de um sítio de ligação diferente dos demais, indicando ser um possível transportador de ácido ferúlico. Os transportadores ABC são membros de uma família protéica que possuem a habilidade de se ligar e hidrolisar ATP como fonte de energia para dirigir o transporte através da membrana plasmática, podendo funcionar como exportadores, importadores ou canais ATP regulados. Desta forma, o objetivo deste trabalho foi analisar a função do gene OsABCB17 como um possível transportador de ferulato em arroz, utilizando dois mutantes oriundos da inserção do transposon TOS17, por meio de análises moleculares e fisiológicas. Esses mutantes apresentaram aumento do teor da lignina na bainha fonte e na folha dreno, sem alterações em outros orgãos, porém a expressão dos genes analisados foi alterada negativamente em alguns orgãos. Em experimentos de pertubação da homeostase do metabolismo dos fenilpropanóides, por meio da adição exógena de ácido ferúlico e de ácido piperolínico (PIP), inibidor da enzima C4H, observamos que, enquanto em plantas WT houve uma redução da expressão dos genes 4CL2, COMT, CAD2 e PAL em vários orgãos, nos mutantes, ou não houve alteração, ou menos genes foram afetados. Esses dados sugerem que a falta de uma correlação entre o teor de lignina e a expressão dos genes pode ser explicada pela complexidade da via, ou simplesmente pelo fato desse transportador estar envolvido em outro tipo de via, como a da suberina, a qual possui sua formação similar a da lignina. Os efeitos positivos observados no crescimento da parte aérea e radicular nos mutantes, provavelmente são devidos à redução da alocação do carbono na síntese de lignina ou suberina, aumentando assim a disponibilidade de energia para o crescimento. Entretanto, a redução da expressão do gene OsABCB17 teve efeitos negativos, tornando as plantas mais sensíveis ao estresse osmótico, provavelmente devido a uma desidratação resultante da redução do acúmulo de suberina e cutícula nas células epidérmicas e subepidérmicas, ou por meio da redução da viiiabsorção de água e/ou da condutividade do xilema. Porém, outros estudos precisam ser realizados nestas plantas mutantes no intuito de obter evidências diretas para fundamentar as hipóteses levantadas nesse trabalho. Ferulic acid is an intermediate in the synthesis of syringyl and guaiacyl, the most abundant monolignol precursor of lignin. The ferulic acid participates in the synthesis of suberin and cuticle, as well as acts as an intermediary for the synthesis of some flavonoids. Through further analysis in the OsABCB17 mutant, a gene of the ABC family, was verified the existence of a different site of connection indicating to be a possible carrier of ferulic acid. The ABC transporters are members of a protein family with ability to bind and hydrolyze ATP. The ATP is used as source of energy to drive the transport across the plasma membrane and may function as exporters, importers or ATP channels regulated (maybe ATP regulated channel). Thus, the aim of this study was to analyze the function of OsABCB17 gene as a possible ferulate transporter in rice, using two mutants derived from the insertion of the Tos17 transposon, by molecular and physiological analysis. These mutants showed an increase of lignin content in the source sheath and in the sink leaf blade without changes in other organs, the two mutants have an increase of lignin content in but the expression of the analyzed genes was negatively altered in some organs.(este paragrafo esta different do original, mas faz sentido aqui) In experiments of disturbance of metabolic homeostasis of phenylpropanoids, through the addition of exogenous ferulic acid and piperonylic acid (PIP), C4H enzyme inhibitor, it was observed that while in WT plants there was a reduction of the expression of 4CL2, COMT, CAD2 and PAL genes in various organs, in mutants, either no change or fewer genes were affected. The data suggest that the lack of correlation between the lignin content and the expression of the genes can be explained by the complexity of the pathway, or simply the fact that the carrier is involved in other type of pathways, such as the suberin, which formation is similar to the lignin. The positive effects in shoot and roots growth are probably due to the reduction of carbon allocation in lignin and suberin biosynthesis, thereby increasing the availability of energy for growth. However, the reduction of the expression of the OsABCB17 gene had negative effects, making the plant more sensitive to osmotic stress, probably due to dehydration resulting from the reduction of the accumulation of suberin and cuticle in epidermal and subepidermal cells, or due to the reduction in water uptake and/or xxylem hydraulic condutivity. However, other studies are needed in these mutant plants to obtain direct evidence to support the hypotheses raised in this work. Keyworks: Lignin, ABC genes, ferulic acid.
