Título: O efeito da hipertensão arterial pulmonar induzida sobre parâmetros testiculares de ratos Wistar submetidos ao treinamento físico resistido
The effect of induced pulmonary arterial hypertension on testicular parameters of Wistar rats submitted to resistance exercise training

Abstract: A hipertensão arterial pulmonar (HAP) é uma doença grave e pouco conhecida, que acomete milhares de pessoas anualmente, comprometendo a qualidade de vida do indivíduo e podendo levar a óbito. Esta doença se caracteriza por vasoconstrição pulmonar excessiva que afeta principalmente pulmões e coração, sem informação sobre alterações em outros órgãos, como o testículo. Um dos tratamentos para doenças vasculares como HAP é a prática de exercício físico, como o treinamento físico de resistência (TFR). Este trabalho objetivou avaliar se a HAP afeta a organização e funcionalidade testicular e se a prática do TFR protege o testículo contra os possíveis danos causados pela HAP. Ratos Wistar machos (60 dias) foram distribuídos em quatro grupos (n = 8 animais/grupo): animais saudáveis sedentários, animais sedentários com HAP, animais saudáveis submetidos ao TFR e animais com HAP submetidos ao TFR. A HAP foi induzida usando duas injeções de monocrotalina (20 mg/kg ip). Os animais foram eutanasiados após 30 dias de TFR. Os testículos foram removidos e processados para análises histológicas, bioquímicas e enzimáticas. Coletou-se sangue para obtenção de soro e determinação da concentração sérica de testosterona (CEUA número 38/2021). Animais sedentários com HAP apresentaram redução na concentração de testosterona e acúmulo de colesterol no testículo, indicando alterações na esteroidogênese. Os testículos destes animais exibiram redução na atividade de enzimas antioxidantes e elevação na concentração de óxido nítrico, promovendo autofagia e morte celular. Alterações metabólicas e enzimáticas resultaram em desorganização na arquitetura tecidual e comprometimento na produção de espermatozoides. O TFR em animais com HAP melhorou parâmetros previamente alterados pela HAP, como elevação dos níveis de testosterona e óxido nítrico, reduzindo as alterações histológicas testiculares. Todavia, não foi possível reverter satisfatoriamente o nível de autofagia e a queda na produção espermática diária. Em conclusão, nossos resultados evidenciaram um efeito deletério da HAP na funcionalidade e organização testicular. O TRF, por sua vez, se mostrou um tratamento complementar capaz de minimizar os danos testiculares causados pela HAP. Palavras-chave: Hipertensão pulmonar. Atividade física. Epitélio seminífero.
Pulmonary arterial hypertension (PAH) is a serious and little-known disease that affects thousands of people annually, compromising the individual's quality of life and potentially leading to death. This disease is defined by excessive pulmonary vasoconstriction that mainly affects the lungs and heart, without information on changes in other organs, such as the testis. One of the treatments for vascular diseases such as PAH is physical exercise, such as resistance exercise training (RT). This study aimed to evaluate (i) whether PAH affects testicular organization and functionality and (ii) whether the practice of RT protects the testis against possible damage by PAH. Male Wistar rats (60 days old) were divided into four groups (n = 8 animals/group): healthy sedentary animals, sedentary animals with PAH, healthy animals subjected to RT, and animals with PAH subjected to RT. PAH was induced using two injections of monocrotaline (20 mg/kg ip). Animals were euthanized after 30 days of RT. The tests were removed and processed for histological, biochemical, and enzymatic analyses. Blood was collected to obtain serum and determine serum testosterone concentration (CEUA number 38/2021). Sedentary animals with PAH showed a reduction in the concentration of testosterone and accumulation of cholesterol in the testis, indicating alterations in steroidogenesis. The tests of these animals showed a reduction in the activity of antioxidant enzymes and an increase in the concentration of nitric oxide, promoting autophagy and cell death. Metabolic and enzymatic alterations resulted in disorganization in tissue architecture and compromised sperm production. The RT in animals with PAH improved parameters previously altered by PAH, such as increased levels of testosterone and nitric oxide, reducing testicular histological changes. However, it was not possible to satisfactorily reverse the level of autophagy and the drop in daily sperm production. In conclusion, our results showed a deleterious effect of PAH on testicular functionality and organization. RT, in turn, provided a complementary treatment of the minor testicular damage caused by PAH. Keywords: Pulmonary hypertension. Physical exercise. Seminiferous epithelium.
