Título: Avaliação da citotoxicidade e mecânica de células tumorais tratadas com compostos candidatos a quimioterápicos
Evaluation of cytotoxicity and mechanics of tumor cells treated with compounds that are candidates for chemotherapy

Abstract: s neoplasias, originadas em sua maioria, pelo acúmulo de mutações celulares e caracterizadas por um quadro de mitose contínua e desordenada podem dar origem a uma massa tecidual, conhecida como tumor. Por representar uma das principais causas de morte no mundo, a busca por medicamentos seletivos às células tumorais e que promovam menor desenvolvimento de efeitos colaterais no paciente, faz-se primordial. O objetivo deste trabalho foi avaliar a citotoxicidade in vitro de compostos candidatos a quimioterápicos em células normais e tumorais, bem como, a interferência desses compostos na mecânica celular (migração). Neste trabalho foram testados o extrato etanólico de amora, o derivado sintético de sulfonamidas (BHCN) e os derivados de β-dicetonas (curcumina e DPBP) nas linhagens celulares VERO (epitélio normal de rim de macaco), HEK293 (epitélio normal de rim humano embrionário) e HT29 (adenocarcinoma colorretal humano). A viabilidade celular foi mensurada pelo ensaio de MTT, e os valores de IC 10 , IC 50 e índice de seletividade (IS) foram obtidos. Os tratamentos com o extrato etanólico de amora e com o composto curcumina apresentaram IC 50 para as linhagens normais superiores aos obtidos às linhagens tumorais, permitindo avaliar o IS de 13,61 e 14,0, respectivamente. O composto DPBP não matou a célula normal em nenhuma das concentrações testadas, e apresentou baixo valor de IC 50 para a linhagem tumoral, demonstrando elevada seletividade. O tratamento com o composto derivado de sulfonamidas (BHCN) revelou um IC 50 superior para a linhagem tumoral em relação aos valores obtidos com as linhagens normais, não sendo, portanto, seletivo para esse tipo de câncer. Em conjunto, esses resultados apontam o extrato de amora, a curcumina, e o DPBP como promissores fármacos para o tratamento do câncer colorretal (CCR). Além disso, para complementar os dados observados em estudos prévios sobre a atividade antitumoral do composto DPBP no melanoma, testou-se a atividade antimetastática deste composto na linhagem de melanoma murino – B16F10, através do ensaio de cicatrização de feridas (Wound healing). Os resultados demonstraram que o composto DPBP inibe a migração das células B16F10 em ambas as concentrações de IC 50 eIC 90 , e por isso, ele pode ser considerado um excelente candidato a fármaco para o tratamento do melanoma, por exibir grande seletividade para células tumorais ao mesmo tempo que impede a migração celular, evitando o surgimento de metástases em diferentes tecidos do organismo. Neoplasms, originated mostly by the accumulation of cellular mutations and characterized by a picture of continuous and disordered mitosis can give rise to a tissue mass, known as a tumor. These can be considered benign when they remain in the original tissue and malignant when the cells acquire the ability to invade other tissues. In this case, they are also called cancer. As it represents one of the main causes of death in the world, the search for drugs that are selective to tumor cells and that promote less development of side effects in the patient is essential. The aim of this work was to evaluate the in vitro cytotoxicity of candidate compounds for chemotherapy in normal and tumor cells, as well as the interference of these compounds in cell mechanics (migration). In this work, the ethanolic extract of blackberry, the synthetic derivative of sulfonamides (BHCN) and the derivatives of dibenzoylmethanes (curcumin and DPBP) in the cell lines VERO (normal epithelium of monkey kidney), HEK293 (normal epithelium of human embryonic kidney) were tested and HT29 (human colorectal adenocarcinoma). Cell viability was measured by the MTT assay, and the IC 10 , IC 50 and selectivity index (IS) values were obtained. The treatments with the ethanolic extract of blackberry and with the compound curcumin presented IC 50 for the normal lines superior to those obtained to the tumor lines, allowing to evaluate the IS of 13.61 and 14.0, respectively. The DPBP compound did not kill the normal cell at any of the tested concentrations, and presented a low IC 50 value for the tumor line, demonstrating high selectivity. Treatment with the compound derived from sulfonamides (BHCN) revealed a higher IC 50 for the tumor line compared to the values obtained with normal lines, therefore, it is not selective for this type of cancer. Taken together, these results point to blackberry extract, curcumin, and DPBP as promising drugs for the treatment of colorectal cancer (CRC). In addition, to complement the data observed in previous studies on the antitumor activity of the DPBP compound in melanoma, the antimetastatic activity of the compound was tested in the murine melanoma lineage – B16F10, through the wound healing test (Wound healing). The results showed that the DPBP compound inhibits the migration ofB16F10 cells at both concentrations of IC 50 and IC 90 , and therefore, it can be considered an excellent drug candidate for the treatment of melanoma, as it exhibits great selectivity for tumor cells at the same time. time that prevents cell migration, preventing the appearance of metastases in different tissues of the body.
