Título: O acúmulo de alumínio na parte aérea é comum em espécies herbáceas do cerrado?
The accumulation of aluminum in the aerial part is common in herbaceous species of the cerrado?

Abstract: O alumínio (Al) é o metal mais abundante na matriz mineral do solo e o terceiro elemento mais comum da crosta terrestre. A forma livre do alumínio (Al 3+ ), disponível em solos ácidos é tóxica para muitas espécies e responsável por alterar a disponibilidade de diversos nutrientes essenciais no solo e consequentemente reduzir o crescimento e desenvolvimento das plantas. Existem dois mecanismos principais de resistência ao Al nas plantas, relatados principalmente nas plantas cultivadas: exclusão e tolerância. Embora possam atuar simultaneamente, em geral, cada espécie exibe um mecanismo preferencial, sendo menos frequente a tolerância com acúmulo de Al na parte aérea. Entretanto, o conhecimento desses mecanismos de exclusão e tolerância em plantas nativas herbáceas nos solos ácidos do cerrado é ainda incipiente. O objetivo da pesquisa foi identificar o acúmulo de Al em espécies herbáceas e determinar a localização deste elemento em caule e folhas de diferentes espécies a fim de entender melhor a distribuição deste metal nas plantas do cerrado e fornecer subsídios para esclarecer as diferentes estratégias que permitem sua sobrevivência na presença de Al. Além disso, pretendeu-se verificar se há diferenças na anatomia e no status nutricional de espécies acumuladoras e não acumuladoras (exclusoras) de Al. O trabalho foi desenvolvido na FLONA de Paraopeba, onde foram feitas as coletas e fixação do material para posterior processamento e confecção de lâminas histológicas. Foram coletadas 33 espécies e feitas análises: de quantificação de nutrientes (Al, Fe, N, P, K, Ca e Mg) e identificação dos sítios de acúmulo de Al, na parte aérea, mediante testes histoquímicos (em microscopia de luz). Em 7 espécies foi realizada a microanálise de Raio-X (microscopia eletrônica de varredura), , para a verificação da abundância relativa de Al, Si, Fe, Mn, Ca e Mg. Foi constatado que a maioria das espécies analisadas é acumuladora de Al, segundo a quantificação química, e os testes histoquímicos mostraram que os principais sítios de acúmulo de Al foram as paredes celulares da epiderme, do parênquima clorofiliano e do colênquima, o conteúdo celular de tricomas, os cloroplastos e os núcleos das células. O teste para Al com chrome azurol S mostrou-se muito mais efetivo do que a hematoxilina; a intensidade de coloração dos testes utilizados foi mais intensa no material não incluído em historesina e a intensidade de coloração da reação não reflete a concentração de Al nos tecidos. O EDS e a análise química foram essenciais pois revelaram a presença do Al em espécies onde este elemento não havia sido detectado pelo teste histoquímico. Para melhor interpretação dos resultados o uso das três técnicas foi essencial. Nenhum padrão nutricional típico para acumuladoras de Al e não-acumuladoras foi identificado: o perfil químico é extremamente variável nos dois grupos de plantas. A maioria das espécies possui: epiderme unisseriada com tricomas tectores e/ou glandulares; mesofilo com parênquima paliçádico e esponjoso diferenciados; presença de compostos fenólicos e feixe vascular colateral circundado por esclerênquima. De uma maneira geral, as características anatômicas estão mais associadas às famílias/táxons e não foi possível encontrar um padrão anatômico e nem um perfil químico que torne possível a distinção entre espécies acumuladoras de Al e não-acumuladoras. O acúmulo de alumínio é uma característica marcante entre as espécies herbáceas de diferentes famílias amostradas no cerrado.
Aluminum (Al) is the most abundant metal in the mineral matrix of the soil and the third most common element of the earth's crust. The free form of aluminum (Al3 +) available in acid soils is toxic to many species and responsible for altering the availability of several essential nutrients in the soil and consequently reducing plant growth and development. There are two main mechanisms of resistance to Al in plants, reported mainly in cultivated plants: exclusion and tolerance. Although they can act simultaneously, in general, each species exhibits a preferential mechanism, being less frequent the tolerance with accumulation of Al in the aerial part. However, the knowledge of these mechanisms in native herbaceous plants in cerrado acid soils is still incipient. The objective of the research was to identify the accumulation of Al in herbaceous species and to determine the location of this element in stem and leaves of different species in order to better understand the distribution of this metal in plants of the cerrado and provide subsidies to clarify the different strategies that allow its survival in the presence of Al. In addition, it was sought to verify if there are differences in the anatomy and nutritional status of Al-accumulating and non- accumulating species. The work was developed in the FLONA of Paraopeba, where the samples were collected and fixed for later processing and preparation of histological slides. Thirty-three species were collected and the following analyzes were performed: nutrient quantification (Al, Fe, N, P, K, Ca and Mg); identification of Al accumulation sites in the aerial part by histochemical tests (light microscopy) and X-ray microanalysis (scanning electron microscopy) to verify the relative abundance of Al, Si, Fe, Mn, Ca and Mg. It was found that most of the analyzed species are Al-accumulating according to chemical quantification and histochemical tests showed that the main sites of Al accumulation were the cell walls of the epidermis of the chlorophyll parenchyma and the collenchyma, the cellular content of trichomes and the chloroplasts and the nuclei of the cells. The test for chrome azurol S was shown to be much more effective than hematoxylin; the staining intensity of the tests used was better in the material not embedded in historesin and the color intensity of the reaction does not reflect the concentration of Al in the tissues. EDS and chemical analysis were essential because they revealed the presence of Al in species where this element had not been detected by the histochemical test. For better interpretation of the results, the use of the three techniques was essential. No typical nutritional status for Al-accumulators and non-accumulators was identified; in both groups the chemical profile of the species was extremely variable. Most species have uniseriate epidermis with tector and/or glandular trichomes; phenolic compounds; palisade and spongy parenchyma, and collateral vascular bundle surrounded by sclerenchyma. In general, the anatomical characteristics are more associated to the families / taxa. It was not possible to find neither an anatomical pattern nor a chemical profile that makes possible the distinction between accumulating and non-accumulating species. The accumulation of aluminum is a remarkable characteristic among the herbaceous species of different families in the cerrado.
