Abstract: O presente trabalho visou determinar os melhores solventes na extração dos pigmentos do urucum e os métodos de produção de corantes em pó e em pasta. Ainda, foram avaliados métodos de identificação, quantificação e isolamento destes pigmentos, bem como os fatores que influenciam na reação de saponificação da bixina formando a norbixina e seu sal. Na avaliação dos solventes de extração, foram utilizadas soluções aquosas de KOH 0,1 N e NH4OH 0,5 N, e soluções de NH4OH 0,5 N em etanol 60% e etanol comercial. Os extratos obtidos pelos solventes KOH 0,1 N e NH4OH 0,5 N foram submetidos à secagem em spray-dryer , originando corantes em pó. Já os obtidos pelo NH4OH 0,5 N em etanol 60% e por etanol comercial foram secos em estufa, originando corantes em pasta. Os extratos e corantes concentrados foram caracterizados quanto ao pH, teores de umidade, sólidos totais, bixina e/ou norbixina, cinzas, amônia, potássio e colorimetria, pela análise das coordenadas L*, a* e b*. Os corantes foram submetidos a técnicas de identificação, como espectroscopia de infravermelho, espectrofotometria UV/Visível e CLAE, para determinar o perfil dos pigmentos presentes. Estes pigmentos foram quantificados por espectrofotometria e CLAE. Foram produzidos padrões de bixina e norbixina de elevado grau de pureza, baseando-se em métodos existentes na literatura. Na análise dos fatores que influenciam na reação de saponificação de bixina formando a norbixina e seu sal, foram realizados ensaios avaliando a influência: (i) do tempo de extração ao se utilizar os solventes KOH 0,1 N e NH4OH 0,5 N, à temperatura ambiente, de trinta minutos até 24 horas; (ii) do tempo de extração e da concentração dos solventes, utilizando-se KOH e NH4OH a 1%, 2%, 3%, 4% e 5%, por uma e duas horas de extração; (iii) do tempo de extração à temperatura de 70 ºC, utilizando-se KOH 0,1 N e NH4OH 0,5 N, por uma e duas horas de extração e (iv) de diferentes temperaturas de extração, utilizando-se KOH 4% a 25, 50, 60 e 70 ºC, durante uma hora. As amostras foram analisadas por CLAE e por espectrofotometria. Os resultados indicaram, na avaliação dos solventes de extração, que o solvente mais eficiente foi o NH4OH 0,5 N em etanol 60%, possibilitando o menor teor de pigmentos residuais nas sementes. O etanol comercial foi o solvente menos eficiente. Quanto ao teor de pigmentos nos extratos, o etanol comercial também apresentou menor poder extrator, enquanto os outros solventes apresentaram desempenhos semelhantes. Os diferentes solventes influenciaram no aumento da concentração de compostos presentes nos corantes produzidos, como cinzas, compostos nitrogenados ou potássio. As coordenadas colorimétricas L* , a* e b* variaram com o pH e com o tipo de solvente extrator. Pela espectroscopia de infravermelho verificou-se a presença da bixina em todos os corantes. Para o isolamento de bixina e norbixina foram desenvolvidas novas metodologias, eficientes e de baixo custo, se comparadas às tradicionais. Pela CLAE constatou-se que a bixina foi o componente majoritário da extração dos pigmentos de urucum quando se utilizou tanto os solventes aquosos alcalinos quanto os alcoólicos. Na análise dos fatores que influenciam na reação de saponificação da bixina a norbixina e seu sal, para a base NH4OH, o tempo de extração foi o fator que mais influenciou. Para a base KOH, a concentração e a temperatura de extração foram os fatores que mais influenciaram na hidrólise da bixina. Ambas as técnicas de espectrofotometria e de CLAE para quantificação dos pigmentos de urucum forneceram resultados satisfatórios.
This research aimed to determine the best solvents in the extraction of annatto pigments, and the production methods of powdered and pasty colorants. In addition, methods for identification, quantification and isolation of these pigments were evaluated, as well as the factors that influence in the saponification reaction of bixin to form norbixin and its salt. In the evaluation of extraction solvents, the aqueous solutions of KOH 0,1 N and NH4OH 0,5 N, and the solutions of NH4OH 0,5 N in ethanol 60% and commercial ethanol were used. The extracts obtained by the solvents KOH 0,1 N and NH4OH 0,5 N were submitted to spray-drying, yielding powdered colorants. Those obtained by NH4OH 0,5N in ethanol 60% and by commercial ethanol were dried in a tray dehydrator, yielding pasty colorants. The extracts and the concentrated colorants were characterized for pH, moisture content, total solids, bixin and/or norbixin, ash, ammonia, potassium and colorimetry by the analysis of the coordinates L* , a* and b* . The colorants were submitted to identification techniques, as the infrared spectrometry, UV/Visible spectrophotometry and HPLC, in order to determine the profile of the present pigments. These pigments were quantified through spectrophotometry and HPLC. Bixin and norbixin standards of high purity degree were produced, based in methods already existent in the literature. In the analysis of the factors that influence in the saponification reaction of bixin to form norbixin and its salt, experiments were developed in order to evaluate the influence of: (i) extraction time when using the solvents KOH 0,1 N and NH4OH 0,5 N, at room temperature, from 30 minutes to 24 hours; (ii) extraction time and of solvents concentration when using KOH and NH4OH at 1%, 2%, 3%, 4% and 5%, during one and two hours of extraction; (iii) extraction time at 70 ºC, when using KOH 0,1 N and NH4OH 0,5 N, during one and two hours of extraction; (iv) different extraction temperatures, when using KOH 4% at 25, 50, 60 and 70 ºC, during one hour. The samples were analyzed by means of HPLC and spectrophotometry. The results indicated, in the evaluation of extraction solvents, that the most efficient solvent was NH4OH 0,5 N in ethanol 60%, yielding the smallest content of residual pigments in the seeds. The commercial ethanol was the solvent of smallest efficiency. For the pigment content in the extracts, commercial ethanol was also the solvent that showed smaller extraction power, while the other solvents showed similar performances. The different solvents had influence, also, in the increase of concentration of compounds present in the produced colorants, as ash, nitrogenated compounds or potassium. The colorimetric coordinates L* , a* and b* varied with pH, and with the type of extraction solvent. Through infrared spectrometry the presence of bixin in all colorants was verified. For bixin and norbixin isolation, new methodologies were developed. These methodologies, when compared to the traditional ones, were lower-cost and more efficient. By means of HPLC, bixin was the major component from extraction of annatto pigments, when using both the aqueous alkaline and the alcoholic solvents. In the analysis of the factors that influence in the saponification reaction of bixin to form norbixin and its salt, to the alkali NH4OH, the extraction time was the factor that most had influence. To the alkali KOH, the concentration and the extraction temperature were the factors that most influenced in the bixin hydrolysis. Both spectrophotometry and HPLC techniques for quantification of annatto pigments gave satisfactory results.
