dc.description.abstractSementes recalcitrantes como as de seringueira não toleram baixas temperaturas no armazenamento e nem ser secas a baixos teores de umidade, o que dificulta o seu armazenamento. Além disso, há poucos estudos sobre o seu processo de desenvolvimento e maturação. Objetivou-se estudar as alterações bioquímicas, fisiológicas e anatômicas durante o desenvolvimento das sementes de seringueira bem como seu comportamento durante o armazenamento. O estudo foi desenvolvido em duas etapas, a primeira relacionada ao desenvolvimento das sementes, que foi desenvolvido na Fazenda Experimental da Epamig em Oratórios-MG e nos Laboratórios de Anatomia, de Análise de Sementes da Universidade Federal de Viçosa. O desenvolvimento das sementes foi acompanhado pela marcação de flores durante a antese. A coleta dos frutos ocorreu a partir dos 15 dias após a antese (DAA), até a deiscência dos frutos. A cada 15 dias, foram feitas avaliações do tamanho dos frutos e das sementes, matéria seca, grau de umidade, emergência de plântulas, índice de velocidade de emergência, proteínas resistentes ao calor, enzimas do estresse oxidativo e teor de compostos de reserva das sementes. Os dados de germinação e índice de velocidade de emergência foram submetidos à análise de variância, teste de Tukey e regressão. A maturidade fisiológica das sementes de seringueira ocorreu aos 175 DAA, quando se obteve o máximo conteúdo de massa seca. Maiores porcentagem e velocidade de germinação foram obtidas em sementes colhidas aos 175 DDA e 180 DAA (sementes já dispersas). Aos 175 e 180 DAA, houve maior expressão de proteínas resistentes ao calor de baixo peso molecular (smHSP) e maior atividade das enzimas do estresse oxidativo. O maior acúmulo de compostos reserva nas sementes de seringueira ocorreu próximo à dispersão, tanto no endosperma quanto nos cotilédones e eixo embrionário. Foram observadas adaptações anatômicas que contribuem para manutenção de altas taxas metabólicas ao final do desenvolvimento das sementes, o que favoreceu o consumo dos compostos de reserva, fazendo com que o embrião não apresente dormência. Na etapa relacionada ao armazenamento, as sementes foram acondicionadas em embalagem de papel kraft colocadas em sacos plásticos e armazenadas a 10 oC, 20 oC ou 25 oC, tratadas ou não com fungicidas químicos, Tecto 600 (35 g /100Kg semente), em associação ao Captan (75g/ 100Kg sementes) ou com alecrim (20g/Kg sementes), em delineamento fatorial 3x3x5 (tratamento, temperatura e período de armazenamento) . A cada 15 dias, foram realizadas avaliações quanto às alterações físicas, fisiológicas, bioquímicas e anatômicas. Os dados foram submetido a análise de variância, teste de Tukey e regressão, quando necessário. As sementes de seringueira armazenadas em temperatura de 10 oC apresentaram maior viabilidade e crescimento de plântulas do que aquelas armazenadas em temperaturas de 20 oC e 25 oC, possivelmente por apresentarem maior teor de compostos de reserva ao final do armazenamento. A redução na atividade das enzimas do estresse oxidativo afetou negativamente a viabilidade das sementes e o tratamento com fungicida químico apresentou fitotoxidez às sementes. Portanto, as sementes de seringueira utilizadas neste estudo, atingiram o ponto de maturação fisiológica aos 175 dias após antese (DAA) e quando armazenadas a 10 oC apresentaram viabilidade até 75 dias.pt-BR
dc.description.abstractRecalcitrant seeds like rubber tree seeds does not tolerate low temperatures in the storage and neither drying at low moisture content, what becomes difficult the storage of these seeds. There are few studies about the process of development and maturation. The objective was to study the biochemical, physiological and anatomical alterations during the development of rubber tree seeds, besides their behavior during the storage. The study was developed in two phases, the first related to the development of seeds, was developed at Epamig experimental farm in Oratórios-MG and in the Anatomic Analyzes Laboratory and Seeds Analyzes Laboratory at Universidade Federal de Viçosa. The development of seeds was accompanied by the tagging of flowers during the anthesis. The gathering of the fruits occurs from 15 days after anthesis (DAA), until the dehiscence of fruits. Each 15 days, were realized evaluations of fruits and seeds size, moisture content, seedlings emergence, emergence speed index (ESI), heat resistant proteins, enzymes of the oxidative stress and the reserve compounds content. The dates of germination and ESI were submitted to the variance analyzes, Tukey test and regression. The physiological maturity of rubber tree seeds occurs at 175 DAA, when was not obtained the maximum content of dry mass. Higher percentage and speed of emergence were obtained in seeds gathering at 175 DAA and 180 DAA (seeds was already disperse). At 175 days and 180 DAA, there was higher expression of the heat resistant proteins with less molecular weight (smHSP) and higher activity of the enzymes of the oxidative stress. The greater accumulation of reserve compounds in rubber tree seeds occurs next to the dispersion, both in the endosperm and in the cotyledons and embryonic axis. Was observed anatomical adaptations which contributes to the maintenance of high metabolic taxes at the end of the development of seeds, what favored the consumption of reserve compounds, causing the non-dormancy of the embryo. At the step related to the storage, the seeds were conditioned in kraft paper that were placed inside polyethylene bags and stored at 10oC, 20oC or 25oC, treated or not with chemical fungicidesTecto 600 (35/100Kg seeds) in association with Captan (75g/100kg seeds) or with ground rosemary (20g/Kg), in factorial design 3x3x5 (treatment, temperature and storage period). Each 15 days, were realized evaluations about the physical, physiological, biochemical and anatomical alterations. The dates were submitted to the variance analyzes, Tukey test and regression when necessary. The seeds of rubber tree stored in temperature of 10oC presented higher viability and seedling growth than those stored in temperatures of 20oC and 25 oC, possibly because the higher moisture content, contributes to preserve for longer, the reserve compounds at the end of the storage. The reduction in the activity of the enzymes of the oxidative stress affected negatively the viability of seeds and the treatment with chemical fungicide presented phytotoxicity to the seeds. Therefore, the rubber tree seeds used in this study, reached the physiological point maturity at 175 days after anthesis DAA and when stored at 10oC presented viability until 75 days.en
