Título: Resposta pós-colheita à injúria por frio de três cultivares de manjericão (Ocimum basilicum L.)
Postharvest responses to the chilling injury in three cultivars of sweet basil (Ocimum basilicum L.)

Abstract: O manjericão tem sido utilizado como condimento culinário, planta medicinal popular e como material de fila pela indústria farmacêutica. Apesar da importância do manjericão como produto fresco, não há nenhuma informação na literatura relativo à sensibilidade de cultivares a injúria por frio. O presente trabalho teve como objetivo investigar a diferença entre os cultivares de manjericão Folha Larga, Semi-roxo e Branco armazenados a 5 oC. Foram colhidos ramos terminais de cada cultivar, selecionados e embalados em caixas perfuradas de acetato de celulose ou não embalados e armazenados a 5 oC durante cinco dias. A perda de matéria fresca total dos ramos embalados em caixas de acetato celulose foi 50,2% para Folha Larga, 56,8% para Semi- roxo e 54,3% para Branco, enquanto nos ramos controle foi de 50,4%, 63,3% e 60,4%, respectivamente. Nas folhas embaladas em caixas de acetato celulose o aumento do extravasamento de eletrólitos foi 48% para Folha Larga, 47% para Semi-roxo e 54% para Branco, enquanto nas folhas controle o extravasamento teve aumentado de 59%, 58% e 68%, respectivamente. Não houve degradação de clorofila das folhas armazenada a 5 oC. O aumento da descoloração das folhas embaladas foi 17,5%, 15,7% e 15,7% para Folha Larga, Semi-roxo e Branco, respectivamente. Nas folhas de controle, o aumento em descoloração foi 19,3%, 24,3% e 23,8%, respectivamente. A respiração aumentou até o quarto dia de armazenamento, com aumento da produção de CO 2 de 62,6% para Folha Larga, 48% para Semi-roxo e 82,6% para Branco. Para o tratamento controle a respiração aumentou para 23,2%, 21,9% e 50,9%, respectivamente. A maior produção de etileno para os ramos armazenados foi observada entre o segundo e terceiro dia de armazenamento, com evoluções de 52,3 nL kg -1 h -1 para Folha Larga, 127,3 nL kg -1 h -1 para Semi-roxo e 159,6 nL kg -1 h -1 para o Branco. No tratamento controle a produção máxima de etileno foi 26,6, 95,7 e 65,8 nL kg -1 h -1 , respectivamente. Géis de amido revelaram uma única isoforma de catalase, tanto nos tratamentos expostos ao frio ou não em todos os cultivares. A atividade de catalase nas folhas embaladas aumentou para 53%, 46% e 63% para Folha Larga, Semi-roxo e Branco, respectivamente. Nas folhas controle, o aumento na atividade da catalase foi 61%, 74% e 63%, respectivamente. O cultivar Folha larga foi o mais resistente à perda de matéria fresca. O aumento do extravasamento de eletrólitos e a atividade da catalase foram coincidentes com o desenvolvimento dos sintomas de injúria por frio (descoloração). As mudanças no extravasamento de eletrólito não estão relacionadas à maior sensibilidade dos cultivares a injúria por frio. A maior taxa de respiração foi observada no cultivar Branco, enquanto no cultivar Folha Larga houve a menor produção de etileno ao longo do armazenamento. O cultivar Folha Larga foi o mais resistente no desenvolvimento do sintoma de descoloração ao longo do armazenamento a 5 oC.
Sweet basil has been used as culinary condiment, popular medicinal plant and as row material by the pharmaceutical industry. Despite of sweet basil importance as fresh product, there is no information in the literature regarding the sensitivity of cultivars to chilling injury. The present work had the goal to investigate such differences among the cultivars Folha Larga, Semi-roxo and Branco of sweet basil leaves stored at 5 o C. Terminal stems of each cultivar were harvested, selected, packed in perforated boxes of cellulose acetate or unpacked and stored at 5 o C for five days. The total weight loss of stems packed in cellulose acetate boxes was 50.2% for ‘Folha Larga’, 56.8% for ‘Semi-roxo’ and 54.3% for ‘Branco’, while in the control leaves was increased by 50.4%, 63.3% and 60.4%, respectively. In the leaves packed in cellulose acetate boxes the increase in leakage was 48% for ‘Folha Larga’, 47% for ‘Semi-roxo’ and 54% for ‘Branco’, while in the control leaves the leakage was increased by 59%, 58% and 68%, respectively. No chlorophyll degradation was detected on the leaves stored at 5 o C. The discoloration during storage increased by 17.5%, 15.7% and 15.7% in the boxed leaves of ‘Folha Larga’, ‘Semi-roxo’ and ‘Branco’, respectively. In the control leaves, the increase in discoloration was 19.3%, 24.3% and 23.8%, respectively. The respiration increased until the fourth day of storage, enhancing the CO 2 production by 62.6% for ‘Folha Larga’, 48% for ‘Semi-roxo’ and 82.6% for ‘Branco’. For the control treatment the respiration increased by 23.2%, 21.9% and 50.9%, respectively. The highest ethylene production for the boxed stems were observed between the second and third day of storage, with evolutions of 52.3 nL kg -1 h -1 for ‘Folha Larga’, 127.3 nL kg -1 h -1 for ‘Semi-roxo’ and 159.6 nL kg -1 h -1 for the ‘Branco’. In the control treatment the maximum ethylene production was 26.6, 95.7 and 65.8 L kg -1 h -1 , respectively. Starch gels revealed a single isoform of catalase, in both, chilling injured and not injured leaves in all cultivars. The catalase activity increased by 53%, 46% and 63% in the leaves of boxed ‘Folha Larga’, ‘Semi-roxo’ and ‘Branco’, respectively. In the control leaves, the increase in catalase activity was 61%, 74% and 63%, respectively. The cultivar ‘Folha Larga’ was the most resistant to the weight loss. The increase in the leakage of electrolytes and catalase activity were coincident with the development of chilling injury symptoms (discoloration). The changes in electrolyte leakage were not related to the sensitivity of the cultivar to chilling injury. The highest rate of respiration was observed in the cultivar Branco, while the cultivar Folha Larga had the lowest production of ethylene throughout the storage. The cultivar ‘Folha Larga’ was the most resistant in developing discoloration symptoms throughout storage at 5 o C.
