Título: Avaliação da eficiência de compostos quaternário de amônio na sanitização de tomate, maçã, e rúcula
Evaluation of the efficiency of quaternary ammonium compounds in sanitization of tomato, apple and rocket

Abstract: Compostos de quaternário de amônio possuem propriedades tensoativas, bactericida e fungicida, que podem atuar sinergicamente no processo de sanitização. Objetivou-se avaliar o efeito antibacteriano e antifúngico de cinco compostos quaternário de amôniosobre as bactérias Staphylococcus aureus e Salmonella Enteritidis in vitro e em superfícies vegetais, e defungos filamentosos Penicillium expansum, Penicillium digitatum e Botrytis cinerea in vitro. Especificamente, os compostos quaternários de amônio foram avaliados na sua capacidade de a) alterar as propriedades termodinâmicas da superfície de frutos de tomate e maçã e de folhas de rúcula, e da adesão de S. Enteritidis e S. aureus, b) atuar como sanitizante de bacterianas aderidas nas superfícies de frutos de tomate e folhas de rúcula, e, c) influenciar nas propriedades físico- químicas e qualidade microbiológica de frutos de tomate sanitizados, durante a vida de prateleira. O efeito bactericida e fungicida in vitrode soluções de quaternário de amônio cloreto de alquilbenzil dimetilamônio (BAC), cloreto de metil trialquil amônio (CMTA), cloreto benzil dimetil amônio (CBDMA), metil dodecil benzil trimetil cloreto de amônio (CMBTA) e de um formulado comercial contendo (BAC+etilbenzalcônio) foram avaliadas por macrodiluição e difusão em meio sólido, determinando as suas concentraçoes inibitorias minimas (CIMs). A alteração das propriedades termodinâmicas das superfícies vegetais pré-tratadas com asconcentracoes predeterminadas de BAC e de BAC+ETB no teste in vitro, foi realizado pelo método do ângulo de contato e o uso das equações da termodinâmica de Young e Laplace. As mudanças na adesão de S. Enteritidis e S. aureus nas superfícies de tomate, maça e rúcula foram quantificadas por contagem em placa, e os resultados expressados em Log de UFC/cm2. A capacidade sanitizante foi avaliada após adesão bacteriana em tomate e rúcula por imersão em soluções de BAC e BAC+ETB. As células sobreviventes foram recuperadas e quantificadas por contagem em placa, e os resultados expressos em reduções decimais (RD). A avaliação da alteração das propriedades físico-químicas, cor, sólidos solúveis totais, acidez titulável, firmeza de frutos de tomate foram realizados a cada três dias após sanitização com soluções de BAC, detergente comercial (DET), dicloroisocianurato de sódio (DCICNA), e água como controle. A qualidade microbiológica foi avaliada no dia 0 e 21 do período de conservação. Os frutos foram conservados em bandejas de isopor cobertas com filme PVC a 7°C. Os resultados do teste in vitro verificou a capacidade bactericida dos cinco quaternários de amônio, em determinadas concentrações cada. BAC e BAC+ETB apresentaram também atividade fungicida em todas nas 10 diluições avaliadas. As concentrações inibitórias mínimas CIMs foram determinadas para ambas, S. aureus e S. Enteritidis em 0,03 mg/mL para BAC e 3,12 mg/mL para BAC+ETB. Os amônios quaternários com maior atividade antifúngica, para Penicillium expansum, Penicillium digitatum e Botrytis cinerea, foram BAC, na concentração de 4 mg/L, e o BAC+ETB, na concentração de 100 mg/mL. As superfícies de tomate, maçã e rúcula naturalmente hidrofóbicas mudaram para hidrofílicas quando pré-tratadas com BAC e de BAC+ETB tornando as superfícies termodinamicamente desfavoráveis para a adesão (∆Gadesão> 0) de S. aureus e S. Enteritidis. O pré-tratamento de tomate e rúcula com 125 mg/L de BAC resultou, respectivamente, em contagens de 5,2 log UFC/cm2e 5,1log UFC/cm2de S. aureus.Já, o pré-tratamento com 6250 mg/L de BAC+ETB resultou em 0,16 log UFC/cm2 para tomate e 0,05 log UFC/cm2para rúcula. As contagens de S. Enteritidis nas superfícies de tomate e rúcula pré-tratadas com 125 mg/L de BAC foram 5,0 e 7,8 log UFC/cm2, respectivamente. Estas mesmas superfícies pré-tratadas com 6250 mg/L de BAC+ETB, tiveram contagens de S. Enteritidis de 3,55log UFC/cm2 sobre tomate e 3,8 logUFC/cm2 para rúcula. Os mesmos compostos aplicados como sanitizantes sobre as superfícies vegetais previamente contaminadasatingiram reduções decimais (RD) de 4,8; 6,1; 5,6 para: S. Enteritidis na superfície de maçã, tomate, e rúcula, respectivamente. A sanitização com 6250 mg/L de BAC +ETB, para S. aureus apresentaram 3,6 RD para maçã e 1,0 RD para tomate. A sanitização com 125 mg/L de BAC resultou reduções para S. aureusde 1,4 RD em maçã, 2,8RD em tomate e 2,4RD em rúcula.Com relação as características físico- quimicas, observou-se que a acidez titulável (ATT) foi diminuída(P<0,05) por DET e DCICNA ao longo do período de conservação, com relação aos frutos tratados com água. A relação Brix/ATT não apresentou diferenças entre os tratamentos. A relação a*/b* da escala de cor de Hunter aumentou até o final do período de conservação, e verificando-se aintensificação da cor vermelha em frutos tratados com BAC. Não houve influência na mudança da cor dos outros tratamentos. A firmeza no dia 21 de armazenamento foi diminuída em 57 % e 61 % pelos tratamentos DCICNA e DET, respectivamente, com relação do valor do dia zero. BAC reduziu em 46% a firmeza dos frutos, valor semelhante ao controle. A sanitização dos frutos com BAC, DET e DCICNA, no dia zero, reduziu, respectivamente, em 1,9 RD, 0,9 RD e 1,8 RD a população de mesofilos aeróbios e 1,1 RD, 0,4 RD e 0,9 RD a população de fungos filamentosos e leveduras. A persistência da ação dos quaternários de amônio ficou semelhante às do controle (água) no final do período de conservação, verificando-se a sua falta de persistência. Conclui-se que soluções de BAC e de BAC+ETB são alternativas ao uso dos compostos clorados como a DCICNA por reduzir a população bacteriana e desfavorecera adesão bacteriana, sem alteração das propriedades físico-químicas dos vegetais.
Quaternary ammonium compounds have surfactant properties, bactericide and fungicide, which may act synergistically in the sanitization process. This study aimed to evaluate the antibacterial and antifungal effect in vitroof five quaternary ammonium compounds on the bacteria Staphylococcus aureus and Salmonella Enteritidis in vitro and vegetables surfaces, and filamentous fungi Penicillium expansum, Penicillium digitatum and Botrytis cinereain vitro. Specifically, quaternary ammonium compounds were evaluated on their ability to a) change the thermodynamic properties of the tomatoand apple fruit surfaces and rocket leaves, and S. Enteritidis and S. aureusadhesion, b) act as sanitizing of bacteria attached on the surfaces of fruits of tomato and rocket leaves, and, c) influence on the physicochemical properties and microbiological quality of fruits of tomato sanitized, during shelf life. The bactericidal and fungicidal in vitro quaternary solutions of ammonium chloride alquilbenzil dimethyl (BAC), methyl trialkyl ammonium chloride (CMTA) benzyl dimethyl ammonium (CBDMA) chloride, methyl dodecyl benzyl trimethyl ammonium chloride (CMBTA) and a commercial formulated containing (BAC+etilbenzalcônio) were evaluated by determining the minimum inhibitory concentrations (MICs) for macrodilution and broadcast on solid medium. The alteration of the thermodynamic properties of the pretreated vegetable surfaces, with concentrations of BAC and BAC + ETB predetermined in vitro assay was conducted by the contact angle method and the use of thermodynamic equations of Laplace and Young. Changes in the adhesion of S. Enteritidis and S. aureus on surfaces of tomato, apple and rocket were quantified by counting in a plate and the results expressed in Log CFU/cm2. The sanitizing capability was evaluated after bacterial adhesion in tomato and rocket dip in BAC solutions and BAC + ETB. The surviving cells were recovered and quantified by counting in a plate and the results expressed as decimal reduction (DR). Assessment of the alterations in physico-chemical properties, color, soluble solids, titratable acidity, firmness of tomato fruits were performed every three days after sanitization with BAC solutions, commercial detergent (DET), sodium dichloroisocyanurate (DCICNA) and water as control. The microbiological quality was evaluated on day 0 and 21 of the conservation period. The fruits were stored in covered Styrofoam trays with plastic wrap to 7 °C. The in vitro assay results verified the bactericidal capacity of the five quaternary ammonium coumpounds at certain concentrations each. BAC and BAC + ETB also showed fungicidal activity in all the 10 evaluated dilutions. The minimum inhibitory concentrations (MICs) were determined for both, S. aureus and S. Enteritidis at 0.03 mg/ml for BAC and 3.12 mg/ml for BAC + ETB. The quaternary ammoniums with higher antifungal activity to Penicillium expansum, Penicillium digitatum and Botrytis cinerea were 4000 mg/l of BAC and BAC + ETB at concentration of 100 mg/ml. The surfaces of tomato, apple and rocket, naturally hydrophobic, changed to hydrophilic when pretreated with BAC and BAC + ETB, making thermodynamically unfavorable surfaces for adhesion (∆Gadesão> 0) of S. aureus and S. Enteritidis. Pretreatment of tomatoes and rocket with 125 mg/L BAC resulted, respectively, in counts of 5.2 log CFU/cm2 and 5.1 log CFU/cm2 of S. aureus. Allready, the pre-treatment with 6250 mg/L BAC +ETB resulted in 0.16 log CFU/cm2 for tomato and 0.05 log CFU/cm2 for rocket. The S. Enteritidis counts the surfaces of tomato and rocket pretreated with 125 mg/L BAC were 5.0 and 7.8 log CFU/cm2, respectively. These same surfaces pre-treated with 6250 mg/L BAC + ETB had counts of S. Enteritidis of 3.55 log CFU/cm2 on tomato and 3.8 log cfu/cm2 for rocket. The same compounds applied as sanitizers on the previously contaminated surfaces reached decimal reductions (DR) of 4.8; 6.1; 5.6 to S. Enteritidis on the surface of apple, tomato, and rocket, respectively. The sanitization with 6250 mg/L BAC + ETB, S. aureus showed 3.6 RD and 1.0RD for apple and tomato. The sanitization with 125 mg/L BAC resulted for S. aureus 1.4 RD in apple, 2,8RD in tomato and 2,4RD in rocket. Regarding the physical and chemical characteristics, it was observed that the titratable acidity (TTA) was decreased (P<0.05) for DET and DCICNA throughout the storage period, in relation to the fruits treated with water. The ratio Brix / TTA showed no differences between treatments. The a*/b* Hunter color scale increased until the end of the storage period, and verifying the intensification of red color in fruit treated with BAC. There was no influence in changing the color of the other treatments. Firmness in day 21 of the storage period was decreased by 57% and 61% by DCICNA and DET treatments, respectively, on the day zero value. BAC reduced by 46% the fruit firmness, value similar to the control.The sanitization of fruits with BAC, DET and DCICNA on day zero, reduced respectively by 1.9 RD, RD 0.9 and 1.8 RD population of aerobic mesophilic, and 1.1 RD, RD and 0.4 0.9 RD population of filamentous fungi and yeast. The persistence of action of quaternary ammonium were similar to the control (water) at the end of the retention period, verifying their lack of persistence. We conclude that BAC solutions and ETB + BAC are alternatives to the use of chlorinated compounds such as DCICNA by reducing the bacterial population and discourage bacterial adhesion without changing the physicochemical properties of vegetables.
